Dr. Newby was raised in Norfolk, the son of James E. Newby, Sr., the principle of Rosemont Junior High School. After graduating from Booker T. Washington High School, he went on to attend Lincoln University and Howard University Medical School. Dr. Newby practiced internal medicine in Norfolk and served as president of the Norfolk Medical Society. 

Dr. Newby was also an active participant in Norfolk City Government. In 1965, he was appointed to the Norfolk City Planning Commission, where he served as the chairman of the capital improvements subcommittee and the liaison to the Model Cities program. In addition to his medical and civic responsibilities, Dr. Newby was an accomplished musician who played several instruments. After his untimely death from heart cancer, the Tidewater Heart Association established a fellowship in his honor to further the study of heart research. He was married to Claytea Baker Newby. They had one daughter and two sons, Cassandra, Keith, and James II.
